Interferon tau secretion in cattle embryos in vitro fertilized before and after cryopreservation Arq. Bras. Med. Vet. Zootec.
Araújo,M.C.C.; Vale Filho,V.R.; Ferreira,A.M.; Sá,W.F.; Barreto Filho,J.B.; Camargo,L.S.A.; Serapião,R.V.; Silva,M.V.G.B..
The effect of cryopreservation in IFN-tau, from bovine embryos produced in vitro was evaluated. Two treated groups (G1= fresh bovine embryos, n=59 and G2= freezed embryos, n=84) were used to study the effect of cryopreservation on IFN-tau secretion. After reaching the blastocyst phase, the embryos were kept on individual culture for additional period of 7 days. On days 3 and 7 after the beginning of embryos cultivation, samples of the media culture were taken for IFN-tau secretion titration. Oocysts taken from follicles ranging from 3 to 5mm in diameter were obtained from ovaries of females at slaughterhouse. The embryos were frozen, after being dehydrated with ethylene glycol (1.8m), conditioned on 0.5ml palletes and frozen. Frozen embryos secreted lower...

Light and electron microscopy study of late embryonic development in the land snail Limicolaria flammea (Müller) (Pulmonata, Achatinidae) Zoologia
Egonmwan,Rosemary I..
The late stages of embryogenesis in the achatinid land snail Limicolariaflammea (Müller, 1774) were described using light and electron microscopy. Embryos at various stages of development were present in the eggs during the first hour after they were laid, from 4-cell blastulae to morulae and fairly advanced stages. The advanced embryo which was fully developed on the second day bears a long cephalic sac, first to be developed, attached to the embryo and a podocyst which is attached to the foot of the embryo. Both of these structures are reduced in size as embryogenesis progresses until they finally disappear at about the 7th day after the egg was deposited. The embryonic shell was apparent on the second day and spiral coiling was apparent at about day 5....

Molecular evidence of early vertical transmission of Leishmania ( Leishmania ) infantum in a dog Ciência Rural
Oliveira,Vinícius Vasconcelos Gomes de; Ramos,Rafael Antonio Nascimento; Ramos,Carlos Alberto do Nascimento; Guerra,Neurisvan Ramos; Maia,Frederico Celso Lyra; Alves,Leucio Câmara; Silva Junior,Valdemiro Amaro da.
ABSTRACT: Visceral leishmaniasis (VL) is caused by the protozoon Leishmania infantum . Transmission of this parasite to hosts occurs mainly through the bite of infected sand flies. However, alternative infection routes have been hypothesized, especially in areas where the biological vector is absent. The exact time of infection and whether in utero transmission occurs have still not been fully elucidated. This report demonstrates molecular evidence of vertical transmission of L. infantum from a pregnant dog to the embryo. Samples (e.g. vulva, vagina, cervix, uterine body, uterine horn and ovaries) from a female naturally infected by L. infantum and from her embryo were molecularly analyzed by means of qPCR and cPCR followed by DNA sequencing. The...

Morphological development of Corydoras aff. paleatus (Siluriformes, Callichthyidae) and correlation with the emergence of motor and social behaviors Iheringia, Sér. Zool.
Rodríguez-Ithurralde,Daniel; del Puerto,Gabriela; Fernández-Bornia,Fernando.
Here we examine major anatomical characteristics of Corydoras aff. paleatus (Jenyns, 1842) post-hatching development, in parallel with its neurobehavioral evolution. Eleutheroembryonic phase, 4.3-8.8 days post-fertilization (dpf); 4.3-6.4 mm standard length (SL) encompasses from hatching to transition to exogenous feeding. Protopterygiolarval phase (8.9-10.9 dpf; 6.5-6.7 mm SL) goes from feeding transition to the commencement of unpaired fin differentiation, which marks the start of pterygiolarval phase (11-33 dpf; 6.8-10.7 mm SL) defined by appearance of lepidotrichia in the dorsal part of the median finfold. This phase ends with the full detachment and differentiation of unpaired fins, events signaling the commencement of the juvenile period (34-60 dpf;...

Oocyte development in cattle: physiological and genetic aspects R. Bras. Zootec.
Britt,Jack H..
Oocytes in cattle are formed during embryogenesis and develop within individual follicles in the cortex of the ovary. Dormant primordial follicles become active and undergo progressive development at regular intervals commencing during the late fetal stage and continuing throughout adulthood. Once activated, follicles and oocytes in a cohort either grow to maturation and ovulation or undergo atresia, ultimately depleting the ovaries of germ cells. It takes an estimated 100 days for a follicle and its oocyte to reach the mature ovulatory stage. Within an individual, number of follicles in one ovary is similar to number in the other ovary; however, there are large differences among individuals in total number of follicles present in both ovaries. In...

Sprícigo,J.F.W.; Leme,L.O.; Guimarães,A.L.; Oliveira Neto,J.C.; Silva,P.C.P.; Moreira,N.H.; Pivato,I.; Silva,B.D.M.; Ramos,A.F.; Dode,M.A.N..
ABSTRACT Piau porcine blastocysts were submitted to MALDI-TOF to identify the main phospholipids (PL). After that, in vivo blastocysts (D6) were vitrified (n=52), non-vitrified were used as control (n=42). After warming, blastocysts were in vitro cultured to assess re-expansion and hatching at 24 and 48 hours. Finally, at 48 hours, hatched blastocysts were submitted to RT-qPCR searching for BCL2A1, BAK, BAX and CASP3 genes. For MALDI-TOF, the ion intensity was expressed in arbitrary units. Blastocyst development was compared by Qui-square (P< 0.05). Among the most representative PL was the phosphatidylcholine [PC (32:0) + H]+; [PC (34:1) + H]+ and [PC (36:4) + H]+. Beyond the PL, MALDI revealed some triglycerides (TG), including PPL (50:2) + Na+, PPO...

Preimplantation Development of Embryos in Labrador Retrievers OAK
ABE, Yasuyuki; SUWA, Yoshinori; YANAGIMOTO-UETA, Yoshiko; SUZUKI, Hiroshi; 鈴木, 宏志.
Preimplantation development of canine embryos is not well understood. To understand the timing of preattachment embryogenesis relative to the luteinizing hormone (LH) surge, early embryonic development was examined in Labrador Retrievers after artificial insemination. The embryos migrated from the oviduct to the uterus beginning on day 11 after the LH surge. This transport must be completed within 24 h. By day 13 after the LH surge, all of the embryos had moved and were localized in the uterus. The embryos developed to the morula stage within 11-13 days and to the blastocyst stage within 14 days after the LH surge, respectively. These findings add to the current understanding concerning the physiology of preimplantation development and should help further...

Freitas,Vicente J.F.; Serova,Irina A.; Andreeva,Lyudmila E.; Dvoryanchikov,Guennadi A.; Lopes-Jr.,Edilson S.; Teixeira,Dárcio I.A.; Dias,Luciene P.B.; Avelar,Suely R.G.; Moura,Raylene R.; Melo,Luciana M.; Pereira,Alexsandra F.; Cajazeiras,João B.; Andrade,Maria L.L.; Almeida,Karlliely C.; Sousa,Francisco C.; Carvalho,Antonio C.C.; Serov,Oleg L..
In order to produce transgenic goats with hG-CSF, a total of 24 adult Saanen and 48 adult undefined breed goats were used as donors and recipients, respectively. Donors were estrus-synchronized with vaginal sponges and superovulated by a treatment with 200 mg FSH given twice daily in decreasing doses over 3 days starting 48 h before sponge removal. Ovulation was induced by injecting 100µg GnRH 36 h after sponge removal. The recipients also received an estrus synchronization treatment. Donors were mated with fertile Saanen bucks and, approximately 72 h after sponge removal, zygotes were recovered surgically by flushing oviducts. The recovered zygotes were briefly centrifuged to a reliable visualization of the pronuclei. The DNA construct containing hG-CSF...

Aires,M.B.; Santos,J.R.A.; Souza,K.S.; Farias,P.S.; Santos,A.C.V.; Fioretto,E.T.; Maria,D.A..
The function of the visceral yolk sac (VYS) is critical for embryo organogenesis until final fetal development in rats, and can be affected by conditions such as diabetes. In view of the importance of diabetes during pregnancy for maternal and neonatal health, the objective of this study was to assess fetal weight, VYS cell markers, and viability in female Wistar rats (200-250 g) with induced diabetes (alloxan, 37 mg/kg) on the 8th gestational day (gd 8). At gd 15, rats from control (n=5) and diabetic (n=5) groups were anesthetized and laparotomized to remove the uterine horns for weighing of fetuses and collecting the VYS. Flow cytometry was used for characterizing VYS cells, and for determining mitochondrial activity, cell proliferation, DNA ploidy, cell...

Reactive oxygen species in bovine embryo in vitro production Biocell
Dalvit,G.C.; Cetica,P.D.; Pintos,L.N.; Beconi,M.T..
Oxidative modifications of cell components due to the action of reactive oxygen species (ROS) is one of the most potentially damaging processes for proper cell function. However, in the last few years it has been observed that ROS participate in physiological processes. The aim of this work was to determine ROS generation during in vitro production of bovine embryos. Cumulus-oocyte complexes were recovered by aspiration of antral follicles from ovaries obtained from slaughtered cows and cultured in medium 199 for 22 h at 39ºC in 5% CO2: 95% humidified air. In vitro fertilization was carried out in IVF-mSOF with frozenthawed semen in the same culture conditions and embryo in vitro culture in IVC-mSOF at 90% N2: 5% CO2: 5% O2. ROS was determined in denuded...

Relationship between eggplant seed morphology and germination Rev. bras. sementes
Silva,Vanessa Neumann; Cicero,Silvio Moure; Bennett,Mark.
Structural differences such as abnormalities, damage and free spaces in seeds may affect germination. The aim of this study was to study the relationship between eggplant seed morphology and seed germination. Ten seed lots of the eggplant cultivar Embu were evaluated by X-ray image analysis and the germination test. Seed image analysis was performed by Image Pro Plus® software and the whole seed area and free space between the embryo and endosperm were measured. The internal seed area filled by the embryo and endosperm was calculated from the difference between the whole seed and free space areas. Based on these results and visual seed analysis, seeds were classified into three categories and information on germination was obtained for each one. X-ray...

Setting of a procedure for experimental incubation of Pacific oyster (Crassostrea gigas) embryos ArchiMer
Suquet, Marc; Amourda, Christopher; Mingant, Christian; Queau, Isabelle; Lebrun, Luc; Brizard, Raphael.
Standardized experimental protocols designed to study Pacific oyster (Crassostrea gigas) gamete quality have not been previously published. Gamete sampling variations and confounding effect of interacting factors result in large variations between replicates, decreasing the effects of the studied factors. This work aims at defining a standardized procedure for incubation of oyster embryos designed for experimental purposes. In a first phase, four experiments were developed to improve embryo sampling and handling. They showed that a minimum of 50 embryos must be counted to decrease the variation between counting replicates. For reliable results, sampling must be carried out from 0 to 7 min after a careful agitation of seawater containing embryos...

Simultaneous dehydration and infiltration with (2-hydroxyethyl)-methacrylate (HEMA) for lipid preservation in plant tissues Acta Botanica
Oliveira,João Marcelo Santos de.
Although this study aims to develop an improved method for the preservation of reserve lipids in plant tissues for different uses in plant anatomy, it mostly aims to develop an improved method for the identification of lipid reserves where synthesis or storage occurs. The proposed procedures entail only the utilization of (2-hydroxyethyl)-methacrylate (HEMA) as a dehydration agent. One of the procedures is based on the gradual exchange of aqueous HEMA solutions with increasing concentrations. In another procedure, dehydration and infiltration are induced by the presence of silica gel around a modified microcentrifuge tube containing the aqueous HEMA solution with the plant tissues, thus allowing efficient lipid preservation. Both procedures resulted in...

Species-specific differences in diatom-induced anomalies in calanoid copepod embryogenesis: are they linked to the stock-piled antioxidants? IBSS Repository
Khanaychenko, A. N..
Egg production and viability of co-occurring in nature copepods, Calanus helgolandicus and Calanoides carinatus, fed diatom, Thalassiosira rotula, and then switched to dinoflagellate Prorocentrum minimum, were compared in experimental conditions. Egg production of both species on diatom diet was similar (up to 17-22 eggs.female-1.d-1) but no viable nauplii were observed. Development was arrested at different stages depending on degree of anomalies. Abnormal embryos displayed various degrees of deteriorations in pigment distribution and organization of extracellular matrix. Reproductive responses of C.carinatus both to negative (diatom) and positive (dinoflagellate) diets were postponed in comparison with quick responses of C.helgolandicus. After 3 days of...
